Countermeasure

Glycerol works osmotically — it holds water wherever it is. Taken with too little fluid, it draws water from surrounding tissue into the gut, producing cramping, nausea and diarrhoea, and leaving you less hydrated rather than more. 500–750 ml alongside the dose is not optional.

WADA classes glycerol as a plasma expander alongside diuretics, because changing plasma volume alters the concentration of everything else in a urine sample. That makes it a masking agent by definition. Anyone subject to testing should treat this as a genuine risk, not a technicality.

Countermeasure

Worth being clear about: the effect is fuller-looking muscle from fluid retention within it. That is real and visible, and it is not growth. It also means the appearance changes with hydration status day to day, which makes it a poor thing to judge progress by.

Common at higher doses and largely a function of concentration. Splitting the dose and diluting it further usually resolves it. Powder forms buffered onto a carrier are better tolerated than liquid glycerol.

Countermeasure

Deliberately raising body water volume increases the load on the heart and kidneys. Anyone with impaired kidney function, heart failure or uncontrolled blood pressure should not do this.

Overview

Glycerol is the odd one out among pump supplements. Citrulline and arginine work by widening blood vessels; glycerol works by holding water. That difference makes it the more visually dramatic of the two approaches and the only one with a doping-relevant problem attached.

Mechanism of Action

Glycerol is osmotically active — water follows it. Absorbed into the bloodstream, it distributes through body fluids and increases the amount of water they hold, raising both plasma volume and intracellular water in muscle.

The visible result is fuller muscle. The physiological result is a larger fluid reserve, which is why the original research interest was endurance performance in heat: more available water means better thermoregulation and delayed dehydration.

The half-life is short at 30–45 minutes, which is why timing matters more than with most supplements.

What It Is and Is Not

It is a genuine effect. Muscle looks and feels fuller, and there is reasonable evidence for a hydration benefit in endurance work under heat stress.

It is water. Nothing about it builds tissue, and the effect disappears when you stop. That is fine as long as it is understood — the problem arises when the fuller appearance gets read as progress, which then makes normal hydration fluctuations look like losing gains.

The Doping Problem

This deserves its own section because it surprises people.

WADA does not ban glycerol for performance reasons. It bans it as a plasma expander, in the same class as diuretics — because changing plasma volume changes the concentration of everything else being measured in a sample. A substance that dilutes a urine test is a masking agent regardless of intent.

For anyone in tested competition this is not a grey area, and the fact that glycerol is sold in ordinary supplement shops and used as a food additive does not change it.

Where It Sits

Alongside citrulline the mechanisms are complementary rather than redundant: citrulline widens the vessels, glycerol fills them. People combine them for that reason, and it is one of the few supplement stacks with a coherent rationale.

The requirement in both cases is adequate water. With glycerol that is not a recommendation but a condition of it working at all.
